What methodologies are used in reproductive and pollination biology?

As I design experiments for my dissertation on floral evolution, I need to move beyond basic observation. I'm looking for a practical overview of both field and lab techniques from classical to modern that are foundational and essential for rigorous work in this discipline.

In my work, I've seen the field successfully blend classical and modern techniques. For pollination, you start with meticulous field observation, insect collection, and exclusion experiments. In the lab, microscopy for pollen tube growth and floral dissections are fundamental. I would recommend now incorporating molecular tools like DNA barcoding to identify pollen loads or using PCR-based markers to assess paternity and gene flow. The strongest studies often combine ecological observation with genetic verification to tell a complete story. It's this integration that yields the most compelling data.
