Can CuI and TBTA form an effective catalytic complex for click reactions?
I'm synthesizing polymeric networks using CuAAC and need a robust, air-tolerant catalyst. Literature promotes CuI/TBTA, but I've encountered inconsistent reaction rates in my hands. I'm asking to understand the real-world stability and catalytic competency of this specific combination from an experienced practitioner's viewpoint, to troubleshoot my methodology.

In my hands, CuI/TBTA is the workhorse complex for routine CuAAC, primarily because TBTA effectively protects the Cu(I) center from oxidation and disproportionation. I have seen it deliver excellent yields under ambient conditions, especially with added base like DIPEA. However, its effectiveness can depend on solvent purity and the specific azide/alkyne substrates. For highly challenging substrates, I would recommend screening more active ligands like THPTA. For standard applications, it's a reliable and effective choice.
