Southern vs western blotting method
I want to analyse the protein of a given sample. Should I use southern or westering blotting approach for the same?

1. Isolate the protein from the given sample.
2. Place the protein – SDS complex on top of the gel in the well. Also a molecular weight marker should be loaded in the well to determine the molecular weight of proteins.
3. Once the protein samples and markers are loaded, pass the current across the gel.
4. Next the gel is included against the membrane and current is passed across the gel.
5. Next step is immunoblotting.
6. Next the primary antibody needs to be added to the solution to wash away unbound primary antibody. Once this is done add secondary antibody.
7. Once the antibodies are conjugated with enzymes and recognises the primary antibody, wash away unbound secondary antibody.
